白屈菜簡介

白屈菜在中國傳統用藥當中,被用於抗發炎,抗菌,免疫調節,抗癌,利膽,護肝及止痛等功效,目前經體內或體外研究也証實白屈菜確實可能有這些功能存在。西方國家將白屈菜用於治療肝臟疾病,胃潰瘍,口腔感染,止痛,皮疹以及肺結核。白屈菜榨成汁後,也被應用在治療疣,以及角膜混濁[1]。


外觀簡述

莖直立,表皮嫩綠色,附著白粉,多分枝,披白色細長柔毛。莖內含黃色汁液。

多年生草本,高約30-50公分,主根粗壯,成成圓錐形,表皮土黃或暗褐色,鬚根多數。

複葉互生,一至二回單數羽狀分裂,基葉長約10-15公分,小葉5-8對,由上而下漸小,頂端小葉呈倒卵形,上部3裂,每裂又2-3淺裂,基部呈楔形而下延。

花色黃,繖形花序頂生和腋生。萼片2片,橢圓形,疏生柔毛,早落。花瓣4片,倒卵形或長圓狀倒卵形。雄蕊多數,子房條形,由2心皮組成,平滑無毛,花柱短,柱頭頭狀。

果實

蒴果條狀圓柱形,種子細小,成卵圓形,皮外有網狀紋。

使用情況

傳統中草藥主要用於止痛,止咳去痰,利尿解毒,治療慢性氣管炎,百日咳,胃炎,胃潰瘍,腹痛,常言,腸炎,痢疾,黃疸,水腫等症狀[3] 。

活性研究

  • 1. 抗發炎:白屈菜甲醇萃取物餵食以collagen誘導關節炎的DBA/1J小鼠,連續4週給予40及400 mg/kg/day,發現脾臟及淋巴結的TNF-α,IL-6,IFN-γ及B細胞量有出現下降情況。小鼠膝關節的關節炎惡化情況有趨緩現象,並且膝部軟骨的侵蝕現象也有也有顯著的減少。而由CD4+及CD25+調節的T細胞量則呈現增加情況,但血清中的IgG及IgM量下降。
  • 2. 抗菌,抗病毒,抗真菌:根部和地上部位提取部分,可抑制細菌,包括Bacillus cereus,Staphylococcus aureus,Streptococcus mutans,Staphylococci,methicillin-resistant Staphylococcus aureus,multiresistant enterococci,gram-positive bacteria, particularly Bacillus anthracis and staphylococci
。病毒部分包括human immunodeficiency virus 1 (HIV-1),herpesvirus, poxvirus, grippevirus。真菌部分為Candida albicans,Fusarium oxysporum,Botrytis cinerea等。
  • 3. 免疫調節:由白屈菜萃取出的一種蛋白結合多醣類,可促進脾臟細胞,骨髓細胞的有絲分裂,並且增加顆粒性巨噬群落生成細胞(granulocyte macrophage-colony forming cells)的數量。併用重組的IFN-γ,會顯著的誘導NO及TNF-α在老鼠的周邊巨噬細胞產生。

    • 3. chelerythrine IV注射,雄性小鼠,LD50 = 18.5 mg/kg
    • 4. sanguinarine IV注射,雄性小鼠,LD50 = 15.9 mg/kg
    • 5. chelerythrine SC注射,雌性小鼠,LD50 = 95 mg/kg

  • 肝臟毒性
    • 1. 70%乙醇萃取白屈菜地上部位,以1..5及3 g/kg/day口服餵食2及4週後,Wistar大鼠的體重,進食量,酵素活性,肝臟組織切片及丙二醛(MDA)均無顯著增加,而GSH及SOD活性輕微但顯著的下降,特別是在3g/kg/day的劑量下[20]。
    • 2. 副腹腔注射雄性ICR大鼠sanguinarine(1, 5, 10 mg/kg),並在3, 6, 9, 12, 18, 24, 36, 48小時時抽血檢驗。AST, ALT, LDH, 肝臟空泡(hepatic vacuolization), 脂質堆積及氧化呈現增加狀況,10 mg/kg組中,三酸甘油脂下降。細胞(primary hepatocytes)實驗指出細胞死亡及DNA斷裂現象呈現劑量(0-30 μM)及時間(0-24小時)相關性,且過氧化物(ROS)含量明顯上升。在高濃度下,sanguinarine也會抑制粒線體的呼吸功能[16]。
    • 3. ssanguinarine及chelerythrine以3比1比例,2 mg/kg/day及100 mg/kg/day給予豬隻連續90天,經檢測高劑量的sanguinarin/chelerythrine在牙齦及肝臟的殘留量分別為0.55 μg/g及0.15 μg/g。血漿中的sanguinarine濃度則為0.11 μg/ml。在生化,血液,組織切片檢測上均無均異常[18]。

  • 生殖毒性
    • 分別於倉鼠及大鼠懷孕的6-11天及6-15天,肌肉注射Ukrain 0.1, 1.67, 28 mg/kg/day,並無發現任何臨床的中毒症狀,也沒有畸形胎出現。懷孕大鼠的黃體數,胚胎著床位置,著床前後損失量,胎兒存活量,胎盤重,胎兒重及冠臀長(crown-rump length)也都沒有異常。而懷孕倉鼠在胚胎著床後損失量有小幅度的增加,且每胎的胎兒數量也有些微的下降[19]。

  • 基因毒性
    • 1. 給予豬隻sanguinarin/chelerythrine (3:1)混合物,2 mg/kg/day及100 mg/kg/day連續90天,並無在肝臟檢測到任何DNA-adduct產物[18]。
    • 2. 0.8 μg/ml sanguinarine及2.0 μg/ml chilidonine給予human acute T- lymphoblastic leukaemia MT-4細胞,以comet assay測試僅發現sanguinarine有產生DNA傷害,而chelidonine則無[17]。

  • 細胞毒性
    • 1. 0.8 μg/ml sanguinarine及2.0 μg/ml chilidonine給予human acute T-lymphoblastic leukaemia MT-4細胞,測得caspase 3及9,Bax的活性及含量上升,細胞質中的cytochrome c含量也明顯提高。Chelidonine也檢測到會使MT-4細胞靜止於細胞週期的G2/M期[17]。

  • 神經毒性
    • 1. Chelidonine對中樞神經系統可產生抑制作用,引起昏睡,血管運動中樞痲痹。若直接服用新鮮植株,則可能產生強烈的胃腸道刺激作用[3]。
